  • Venetoclax, also known as ABT-199, is a BCL-2-selective inhibitor. It blocks the growth of BCL-2-dependent tumors in vivo and spares human platelets. A single dose of venetoclax in three patients with refractory chronic lymphocytic leukemia induced tumor lysis within 24 hours. Souers A.J., et al. "ABT-199, a potent and selective BCL-2 inhibitor, achieves antitumor activity while sparing platelets." Nat. Med. 19: 202-208 (2013).
  • The high sensitivity to venetoclax was dependent on human multiple myeloma cell lines (HMCLs) with the CCND1 translocation. Six out of eight CCND1 cell lines were efficiently inhibited by venetoclax with a median LD50 value of 10 nM. Of note, the median venetoclax LD50 of sensitive HMCLs (10 nM) was significantly lower than that of peripheral blood lymphocytes from healthy donors (200 nM). Touzeau C., et al.   • Both the BH3 mimetic venetoclax and phosphatidylinositol 3-kinase (PI3K)/mammalian target of rapamycin (mTOR) inhibitors improved tumor responses to the antiestrogen tamoxifen in estrogen receptor-positive breast cancer.
