- Binds specifically to heat shock protein HSP90 and its GP96 homologue; inhibits protein maturation. Whitesell, L. and Cook, P. "Stable and specific binding of heat shock protein 90 by geldanamycin disrupts glucocorticoid receptor function in intact cells." Mol. Endocr. 10: 705-712 (1996).
- Stability: Geldanamycin's methoxy group on the quinone ring is reactive -- it can be displaced by other alcohols or water. Thus, geldanamycin is unstable in water and ethanol, and those solvents should not be used for making stock solutions. Although geldanamycin stability in methanol would not be a problem (because no structural change occurs if the geldanamycin reacts with methanol -- it is just a methoxy interchange), solubility in methanol is very poor. DMSO itself probably does not react with geldanamycin, but DMSO can contain moisture from the air and/or small amounts of DMSO degradation products that might react with geldanamycin. Thus, DMSO stock solutions should be kept out of contact with air to the greatest extent possible.
- Solubility in Water: Geldanamycin is very poorly soluble in water; maximum solubility in plain water is estimated to be about 20-50 uM; buffers, serum or other additives may increase or decrease the aqueous solubility. Although geldanamycin is somewhat unstable in water, it is of course unavoidable that biological testing of geldanamycin will expose the compound to water. We do not have any data on the actual rate of geldanamycin decomposition in water or other aqueous media.
